Medium relevanceA NOVEL PHELLINUS STRAIN AND THE METHOD FOR ISOLATING beta-1,3 GLUCOMANNAN THEREFROM SHOWING IMMUNE ENHANCING ACTIVITY AND ANTITUMOR ACTIVITY
Phellinus linteus
Key points
- A beta-1,3 glucomannan polysaccharide isolated from a novel Phellinus strain is provided to increase the immunity and secure the anticancer effect, thereby being used for a pharmaceutical composition or a health functional food for preventing and treating cancers
- A novel Phellinus strain has a sequence of SEQ ID: NO. 1 and is deposited as a deposition no
- A method for isolating beta-1,3 glucomannan polysaccharide from Phellinus linteus comprises the steps of: (a) after adding 4-6 times of water to the weight of Phellinus linteus fruit body, extracting it for 3-6 days and filtering, dialyzing and freeze-drying the extract to obtain a water-soluble extract (b) after mixing a solid ammonium sulfate with the water soluble extract and leaving it to remove precipitated materials therefrom, filtering it to obtain a purified glycan fraction (c) adding sodium carbonate reagent to the fraction and recovering only acidic glycan fraction therefrom using a column (d) after adding an acetic acid to the acidic glycan fraction to adjust the pH into 4.0-6.0, adding an organic solvent thereto to obtain precipitated materials and (e) subjecting the precipitated materials to Sephadex chromatography using a liquid acetic acid to fraction glycans by size
- A pharmaceutical composition for increasing immunity and preventing and treating cancer comprises the beta-1,3 glucomannan polysaccharide obtained by the method as an effective ingredient

Abstract
A beta-1,3 glucomannan polysaccharide isolated from a novel Phellinus strain is provided to increase the immunity and secure the anticancer effect, thereby being used for a pharmaceutical composition or a health functional food for preventing and treating cancers. A novel Phellinus strain has a sequence of SEQ ID: NO. 1 and is deposited as a deposition no. KCCM KSSW01. A method for isolating beta-1,3 glucomannan polysaccharide from Phellinus linteus comprises the steps of: (a) after adding 4-6 times of water to the weight of Phellinus linteus fruit body, extracting it for 3-6 days and filtering, dialyzing and freeze-drying the extract to obtain a water-soluble extract (b) after mixing a solid ammonium sulfate with the water soluble extract and leaving it to remove precipitated materials therefrom, filtering it to obtain a purified glycan fraction (c) adding sodium carbonate reagent to the fraction and recovering only acidic glycan fraction therefrom using a column (d) after adding an acetic acid to the acidic glycan fraction to adjust the pH into 4.0-6.0, adding an organic solvent thereto to obtain precipitated materials and (e) subjecting the precipitated materials to Sephadex chromatography using a liquid acetic acid to fraction glycans by size. A pharmaceutical composition for increasing immunity and preventing and treating cancer comprises the beta-1,3 glucomannan polysaccharide obtained by the method as an effective ingredient.
